Counterion condensation was employed to achieve polyelectrolyte collapse for naturally occurring polymers, hyaluronic acid (HA) and alginic acid (AL). Secondary crosslinking led to the formation of the desired nanoparticles (NPs). HA or AL NPs were stabilized by covalent crosslinker with the help of carbodiimide. Ideal pH ranges for the most extended conformation of HA and AL were pH 5.5-7.5 and 6.5-7.5 respectively. It was found that HA and AL can be condensed with Na+ ions (>0.25 M) and Ag+ ions (>100 mM, HA only). Particle sizes were examined using kinematic viscosity (St), dynamic light scattering (DLS), and atomic force microscopy (AFM). DLS was able to assess the polydispersity of the NPs in the solution, where two main peak distributions for HA, 1-10 nm and 100-1000 nm, and 160-280 nm for AL. AFM results confirmed the presence of multiple particle sizes for HA NPs, with the height between 1-4 nm.
