Tea has been reported to have beneficial health effects in humans. While experimental studies have supported the chemopreventive activity of tea, the reported effects in humans are less clear. This dissertation examined the effects of green tea on oxidative stress (study 1) and working memory (study 2) in humans. In the first study, the effect of green tea (as an ice tea drink) on oxidative stress biomarkers in smokers and non-smokers was examined using a double-blind, placebo-controlled, crossover design. Smokers (10 women, 2 men) and non-smokers (9 women, 6 men) consumed green tea (2.75%, 32 oz/day) or placebo for a week. Blood and urine samples were analyzed for oxidative stress biomarkers by High Performance Liquid Chromatography (HPLC). Urinary and plasma malondialdehyde (MDA) in smokers were higher than that seen in non-smokers. After green tea, urinary MDA, urinary 8-hydroxy-2' -deoxyguanosine (8-OHdG), and 8-OHdG in white blood cells (WBC) were decreased by 65.7%, 43.2% and 42.8%, respectively. Urinary MDA and WBC 8-OHdG in subjects with high basal oxidative stress were significantly reduced by 53.9% and 53.5%, respectively, after green tea compared to placebo (P < 0.05). In a second study, the effect of green tea extract (capsules) on working memory was examined using a single-blind, placebo-controlled, crossover design. Twenty non-smoking Caucasian women were recruited and placed in two age groups (< 30 y and 50 - 70 y). Subjects received green tea extract (900 mg x 6) or placebo over 24 hours. Working memory was measured by reading span, operation span, and N-back task paradigm (Inquisit 4.0.4, Millisecond Software, Seattle, WA). Green tea extract significantly improved reading span performance and reduced 1-back latency in older women, but not in the younger women. In conclusion, green tea significantly reduced oxidative DNA damage and lipid peroxidation in smokers and non-smokers and improved some aspects of working memory function in older women.
