Investigation of local chemical fluxes is of great significance in a wide range of research areas, including neuroscience, cell biology, and electrochemistry as well as material science. Electrochemical detection methods have emerged as an increasingly important means for measurements of local chemical fluxes due to high sensitivity and specificity and superb spatio-temporal resolution when coupled with scanning probe techniques. Nanopipettes (i.e., pipettes with tip opening on the scale of tens to hundreds of nanometers) have been developed and employed in a variety of applications related to local chemical analysis, mostly notable as nanoscale delivery tools or scanning probes in scanning probe techniques such as scanning ion conductance microscopy (SICM).In this thesis, with the aim to extend the utilities of nanopipettes, specifically, to improve quantitative aspects of nanopipette delivery and to incorporate chemical specificity into SICM imaging, several unconventional nanopipette configurations were developed and employed for a variety of applications. A carbon ring/nanopore electrode was used to investigate the fundamental ion transport mechanism through a nanopipette, and the influence of surface charge-induced concentration polarization effects on ion delivery for small pipettes was studied. Next, a gold crescent/nanopore electrode was developed and used for hybrid scanning ion conductance-scanning electrochemical microscopy (SICM-SECM) to incorporate chemical specificity into conventional SICM topographical imaging, and heterogeneity in permeability of degraded Nafion membranes was examined. Furthermore, dual-barrel membrane patch-ion channel probes (MP-ICPs) were developed and applications of MP-ICPs in localized chemical sensing with high spatial resolution were demonstrated.

The ultimate goal of these endeavors is to develop tools and methodologies that have the ability to map and manipulate local chemical fluxes with high spatio-temporal resolution and high sensitivity, and, in turn, to diversify the scope and applications of nanopipette-based analytics.
