Colon cancer is still the second leading cause of cancer deaths in the United States. Epigenetic gene silencing involving DNA Methyltransferases and Historic Deacetylases plays an important role in the progression of colon cancer. We proposed that EGCG inhibits DNMTs and HDACs. Using an in vitro model of colon cancer where key regulatory genes are silenced via methylation, we found that colon cell's sensitivity to methylation plays a role in its response to the green tea polyphenol, EGCG. HDACs and DNMT protein show reduced expression when treated with EGCG in the methylation sensitive HCT 116 human colon cancer cell line but relatively stable expression in the HT-29 cell line. This decreased expression may he partially explained by our finding that DNMT3a and HDAC3 are degraded in the methylation sensitive colon cancer cell line in part by EGCG inhibiting DNMI3a and HDAC3 association with the E3 ubiquitin ligase, UHRF1. These findings provide a rationale for the development of a more targeted therapy for methylation sensitive colon cancers that can include EGCG in combination with other DNMT and HDAC inhibitors.
