Two types of Asian Domestic Buffalo have been recognized, the so-called Swamp Buffalo, which is used as a draft and meat animal, and the River Buffalo, which is used for milk production. The Asiatic buffalo has two subspecies: the river buffalo with a karyotype 2n = 50 and the swamp buffalo (2n = 48). These two subspecies differ in a chromosome pair, but they can be inter-breed. The hybrid has a chromosome number of 2n = 49 and may have a lower reproductive performance due to the potential for the production of incompetent gametes (aneuploidy) and subsequent abnormal embryos which usually die in early embryonic life. Water buffalo are also notorious for their poor superovulatory responses, with 0 to 2 transferable embryos per superovulation/flush commonly reported.

These series of studies allowed determination of differences between riverine type and hybrids in regard to endocrine patterns, follicular development, ovulation rates, and corpus luteum formation during the estrous cycle. The interovulatory interval in water buffaloes ranged from 17 to 23 days regardless of karyotype. This interval was affected by the number of follicular waves observed, with more waves resulting in longer estrous cycles, differences on the second subordinated group of follicles, dominant follicle diameter, ovulatory follicle diameter, size at follicular deviation, and follicle growth rates between buffalo with a normal karyotype and hybrids were found. Circulating concentrations of P4 and estradiol were not different, whereas inhibin concentrations were greater for hybrid than normal karyotype buffalo. Our findings indicate that with the exception of inhibin, the hormonal patterns were similar between karyotypes.

These findings suggest failure to ovulate after the superovulation treatment with FSH in water buffalo, during and out of the favorable reproductive season, although the follicular recruitment and further development of ovulatory follicles was evident despite chromosome number. Also, gonadotropin stimulation causes a distinct pattern of secretion of hormones particularly inhibin in riverine and hybrid buffalo, and thus, it may affect the preovulatory timing of E2 and LH, leading to low spontaneous ovulation rates or outright failure to ovulate, high anovulatory rates and low embryo production following superstimulation with gonadotropins regardless karyotype.
