Urinary hydroxyproline and hydroxyproline:creatinine ratios were used as indicators of exercise-induced collagen degradation.

Twelve healthy male and female (18 to 30 years of age) participants consumed a vegetable and fruit-free diet for 28 days and a collagen-free diet for 6 days (Days 23 to 28). In a double-blind fashion, participants were supplemented with Pycnogenol(TM) (150 mg day-1) or placebo (V-caps) for the study duration. Pre-exercise 24-hr urine collection began on Day 24 and the exercise task (50 eccentric maneuvers of the right quadriceps muscle group at an angular velocity of 30 deg s-1 was performed on Day 25. Post-exercise 24-hr urine was collected on 2 and 3 days following the exercise task (Days 27 and 28). Following eccentric exercise subjective muscle pain or soreness was significantly greater for the placebo condition on post-exercise Day 2 than for Pycnogenol(TM). Muscle pain or soreness was greater for post-exercise Days 2 and 3 for both conditions with the scores significantly greater for the placebo than the Pycnogenol(TM) condition on post-exercise Day 2. Urinary hydroxyproline:creatinine ratios were significantly lower for the Pycnogenol(TM) than for the placebo condition suggesting that there was lower collagen turnover before and following eccentric work for the Pycnogenol(TM) condition.

It was concluded that the administration of Pycnogenol(TM) attenuates the perception of muscle pain or soreness following eccentric work. Pycnogenol(TM) may also lessen the extent of muscular damage through its effects on muscle collagen.
