This study applies the AFM technique to the characterization of the polysaccharide hyaluronan. AFM allows observation of samples at a single molecule level; furthermore, the conditions used for sample preparation for AFM imaging (i.e., deposition on a surface), can influence the chain conformation, and allow detection of forms energetically possible but non observed in solution.

The first part of this study compares the images of low, moderate and high molecular weight hyaluronan obtained after deposition on either a freshly cleaved or a previously cleaved (i.e. prehydrated) mica surface. The polysaccharide exhibits a stronger affinity for the pre-hydrated surface, evident in the high incidence of extended forms. When deposited on a freshly cleaved surface, hyaluronan tends to show self-interactions, aggregation, and less extended conformations, in same cases resembling loose helices. The moderate molecular weight sample forms compactly condensed rods under some conditions.

A characterization of the pre-hydrated mica surface is then presented. The hydration layer is shown to be a structured layer, the water molecules forming an ice-like structure, strong enough to avoid its penetration by the tip when operating in soft tapping. Images of the forming structured water layer are presented, as well as images representing its destruction by the heat generated by the experimental set-up. Statistical analysis of the height of the forming or disappearing layer compares well with data reported in the literature.

Finally, different substrates are used for sample deposition, to reveal how the presence of different functional groups on the surface affects the hyaluronan conformation. Interestingly, the polysaccharide does not establish favorable interactions with aminated surfaces, even when all the amino groups are charged. Instead, it shows a high affinity for the hydrophobic graphite surface.

These observations suggest that the hydrophobic patches present on the polysaccharide chain, where there is a high density of -CH groups, play a major role in its interaction with the substrate. Those patches can cause a higher number of molecules to stick to the substrate with little or no occurrence of condensation and aggregation.
