Abnormal adipogenesis, which is characterized by aberrant proliferation and differentiation of fat cells, leads to obesity. In the past decades, obesity has become an epidemic, and is closely associated with other health problems such as type II diabetes.

Adipogenesis is a complicated process that involves both cell proliferation and differentiation. Efforts have been made to study adipogenesis with the hope of understanding, then controlling fat formation in the human body. Unfortunately, until recently, no effective therapeutics have been developed for prevention or treatment of obesity. In this study we try to understand the molecular basis for adipogenesis and seek ways to control it. The initial observation of an anti-adipogenic effect from the use of extract from Banaba leaves (BE) heralded a chain of intensive studies using mouse 3T3-L1 cell line. Sequential purification of BE in our lab led to the identification of tannic acid (TA) as a group of anti-adipogenic compounds. The anti-adipogenic activity of TA might result from the alteration in the expression of c-fos, c-jun and c-myc proto-oncogenes. Based on the structural features of TA, Penta-O-galloyl-D-Glucose (PGG) was chemically synthesized and shown to have strong anti-adipogenic activity. The positive adipogenic factors in response to adipogenesis inducer cocktail (insulin, isobutylmethylxanthine and dexamethasone) were found not to be inhibited by PGG. In contrast, NF-kappaB, the negative adipogenic factor, was activated by PGG in the early stage of adipogenesis. Thus, we propose that the anti-adipogenic activity of PGG does not involve the down-regulation of the positive factors, but correlates with the activation of negative factors. Further studies showed that PGG also has an anti-lipogenic activity in 3T3-L1 adipocytes and a blood triglyceride reducing activity in genetically obese or diabetic mice. These observations suggest that PGG, used as a novel anti-adipogenic and anti-lipogenic compound, may significantly contribute to the better understanding of adipogenesis, and prevention and treatment of obesity.
