The aims of this research are to develop a new direct-reading method involving attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy and a novel whole-glove method to detect breakthrough and permeation of a pesticide, captan, through glove materials. The results are compared with permeation testing conducted using the American Society for Testing and Materials (ASTM) Method F 739-99a for an aqueous challenge concentration of 217 mg/mL Captan 50-WP and hexane collection solvent. Concurrent analyses using gas chromatography-mass spectrometry are performed for all permeation and surface sampling experiments. A solvent casting method is developed to produce uniform Captan surface films on a disposable nitrile glove for quantification with a portable ATR-FTIR spectrometer at 1735 +/- 5 cm-1. The quantifiable limits are 0.31 - 20.7 mug/cm2 ( r=0.9967; p≤0.05) for the outer glove surface, and 0.55 - 17.5 mug/cm 2 (r=0.9409; p≤0.05) for the inner surface. Relative humidity and temperature did not affect the uncoated gloves at this wavelength. Glove screening using ATR-FTIR is necessary to control for between-glove variation. ATR-FTIR is also used to measure the acrylonitrile (ACN) content of disposable nitrile gloves. A moving robotic hand test system is developed to investigate chemical permeation for a whole-glove system. Hand movement did not significantly affect the permeation of captan through nitrile gloves, but did influence the degradation of glove materials, resulting in penetration and catastrophic glove failures. The 8 h permeation results are 36.8 +/- 27.9 mug/cm 2 for the ASTM F 739-99a method with collection solvent, 0.8 +/- 0.3 mug/cm2 using ATR-FTIR with an ASTM F 739-99a test without collection solvent, and about 0.01 to 0.02 mug/cm2 for the whole-glove permeation tests without movement. The collection solvent played a significant role in eliciting increased permeation. Overall, ATR-FTIR can be used in glove surface exposure assessments and to evaluate the effectiveness of personal protective clothing in the field. The robotic hand simulated normal hand motions, is reliable, and could be used to assess the influence of hand movement on the permeation of non-volatile components through gloves. The whole-glove sampling methods developed in this study can be applied to field studies.
