The deep subsurface biosphere was investigated through collection and analysis of samples from approximately 2 km deep in the gold mines of South Africa. Traditional cultivation techniques were employed for the enrichment and isolation of strictly anaerobic microorganisms including methanogens, iron- and manganese-reducing bacteria, and sulfate-reducing bacteria. Methods were selected to bring indigenous organisms into culture in order to elucidate the microbial ecology of this environment. Enrichment cultures were prepared and maintained for the subsequent isolation of the dominant or most numerous microbes within subsurface communities. Isolates were analyzed phylogenetically to aid in their taxonomy, and their physiologies were studied in order to determine their roles in the deep subsurface habitat.

These data were also used to describe new species of microbes when appropriate. Ten microbial isolates including seven phylogenetically-distinct microbes were discovered during this investigation. Each isolate was generally characterized according to phenotype and by analysis of its 16S rRNA gene sequence. Two of the isolates, a bacterial thermophile and a methanogen, were further characterized in preparation for official proposals of novel taxa. Based upon phylogenetic data, it appeared that both organisms represented novel microbial taxa. Several of the bacterial isolates, which were not further characterized, also were phylogenetically distinct and thus warrant further investigation.
