5 and Cuscuta is a facultatively photosynthetic parasite. The DNA sequence of Cuscuta compacta and Cuscuta gronovii psbA andCuscuta compacta psaA were determined. The psbA sequence identities of two dodder genes is only about 88% compared to those of tobacco. A phylogenetic tree based on the psbA DNA sequences shows that the relationships of 11 higher plants matches the traditional classification very well. The long branch of the psbA phylogenetic tree for the two dodder species shows rapid evolution. The amino acid sequences of the D1 and A1 proteins of dodder show higher identities in the regions which are related to the biological functions of those proteins. Two DNA fragments containing the psbA gene were isolated by screening a genomic library, ligated into a plasmid vector, and sequenced. The result shows that the start codon of dodder psbA is GTG, not ATG. The DNA sequence in the promoter region of psbA is totally different from that of other plants except the Subjects--Topical Terms:
