The amphiphilic alpha-helix is a protein secondary structure containing hydrophobic and hydrophilic surfaces. These surfactant-like properties allow amphiphilic helices to self-assemble in aqueous solution burying hydrophobic surface and exposing the hydrophilic surface to the aqueous solution.

The designed amphiphilic alpha-helix nano-1 can non-covalently attach to the surface of single-walled carbon nanotubes (SWNTs) and increase the solubility of SWNTs in aqueous solution. There are two ways to improve the efficiency of nano-1 solubilization of SWNTs. The first is an inter-molecular modification and the second method involves an intra-molecular modification.

For the first method, we used 1-ethyl-3-[3-dimethlaminopropyl]carbodiimide hydrochloride to cross-link nano-1 molecules around SWNTs. Experiments show that cross-linked nano-1/SWNT dispersions maintain more SWNTs in solution over non cross-linked solutions, and the cross-linked nano-1/SWNTs also exhibit a tendency to self-assemble into extended fiber-like structures.

For the second method, we used methanol to increase the helicity of nano-1 molecules. Experiments indicate that a tremendous increase in SWNT concentration was observed with increasing nano-1 helicity.

In this study, we have also designed a series of surfactant peptides which have 8 residues sequences to test the affinity of different amino acids for SWNTs. The experiments were conducted under the conditions that minimize peptide/peptide interactions. Raman spectroscopy and optical spectroscopy were used to study the peptide/SWNT dispersions and peptide/SWNT interactions are discussed.
