Respiratory syncytial virus (RSV), a human pathogen, can enhance allergic responses to allergens by augmenting IgE production. Environmental allergens may play a role in immune responses to RSV. Bovine RSV (BRSV) infection closely parallels human RSV, thus our lab has used this model to study RSV infections and the impact of allergen exposure on RSV immune responses. This project is concerned with the immune response to BRSV during fungal allergen, Alternaria (Alt), exposure prior to and during infection; the same procedure was applied in a second infection 109 days later. Effector phenotype of memory lymphocytes after primary and secondary BRSV infections were studied with and without Alt. PBMCs were collected and used in BRSV restimulation assays for cytokine analysis. Memory lymphocytes were sorted by their high CD44+ CD25-CD62L-profile and restimulated with BRSV. CD25, IL-4, IL-12, and IFNgamma expression were measured in comparison with non-infected controls, and BRSV specific IgG1, IgG2, and IgE were measured in serum. All BRSV infected animals demonstrated quantifiable clinical scores; however, the Alt/BRSV infected group produced lower scores than mock Alt/BRSV animals during the second infection despite producing higher levels of BRSV specific IL-4 and Alternaria specific IgE and lower IgG1 throughout the study. This study also shows that higher clinical scores in a secondary infection correlate to higher IFNgamma levels prior to infection. This bovine model demonstrates that exposure to environmental allergens is a likely immunomodulatory factor in determining the outcome of successive RSV infections.
