The objective of this study was to determine if the flavonoids catechin, epicatechin and silibinin, when added to caprine semen, had any effect on percent sperm motility and progressive motility post-thaw. Semen samples were collected from five post pubertal goats using electro-ejaculation and aliquoted into three replicates. Egg yolk citrate extender supplemented with 0 (control) 10, 100, 1000, or 10000 muM of either catechin, epicatechin or silibinin was used to extend samples prior to cooling (5°C) and cryopreservation. Semen samples were analyzed using Computer Assisted Sperm Analysis to determine the percent sperm motility and progressive motility post-thaw. An ANOVA and the Student--Newman--Keul's multiple range test were performed on arcsine transformed data to determine differences in post-thaw motilities using flavonoid and dosage as the main effects. Sperm motility of samples treated with 10 muM (31.4 +/- 12.6, mean +/- SD), 100 muM (36.2 +/- 15.9), 1000 muM (31.4 +/- 24.3), and 10000 muM (13.0 +/- 4.1) catechin or 10 muM (25.6 +/- 20.4), 100 muM (30.8 +/- 24.1), 1000 muM (32.2 +/- 21.5), and 10000 muM (11.0 +/- 10.9) epicatechin were not significantly different from the control 0 muM (28.2 +/- 4.2). In addition, the motility of samples treated with 10 muM (24.6 +/- 13.4), 100 muM (32.6 +/- 11.2), 1000 muM (29.2 +/- 18.9) of silibinin were not significantly different than the control, although samples treated with 10000 muM of silibinin (10.2 +/- 9.8) were significantly less than the control (28.2 +/- 4.2; P<0.05). Samples treated with 10 muM (4.6 +/- 5.0), 100 muM (5.4 +/- 5.0), 1000 muM (8.2 +/- 13.4), and 10000 muM (17.2 +/- 10.6) catechin or 10 muM (6.8 +/- 9.8), 100 muM (5.4 +/- 5.0), 1000 muM (4.2 +/- 3.6), and 10000 muM (19.2 +/- 18.1) epicatechin did not have a significantly different percentage of progressively motile sperm than the control (3.4 +/- 1.1). Samples treated with 10 muM (4.8 +/- 7.0), 100 muM (5.6 V 5.6), 1000 muM (3.2 +/- 4.4) of silibinin were not significantly different from the control, although the 10000 muM (44.8 +/- 31.4, P<0.05) silibinin treatment contained significantly greater percentage of progressively motile sperm than the control (3.4 +/- 1.1). These findings demonstrate that the cryoprotective and antioxidant properties of sibibinin may aid in the post-thaw progressive motilities of goat sperm.
