Many environmental factors interact with genetics to alter immune development. This dissertation explores factors in early life that alter the trajectory of immune development, focusing mainly on the broad theme of wildfire smoke exposure in early life. We evaluate the long-term effects of wildfire smoke exposure on innate immune responses to pathogen associated molecular patterns in rhesus macaques. Innate cytokine responses are attenuated in juvenile monkeys that were exposed to wildfire smoke in infancy. Many of the immune parameters evaluated normalize as the smoke-exposed animals mature, but an attenuated IL-6 response to LPS persists. Smoke exposed animals were also at greater risk for having undetectable titers following vaccination. In the last chapter, we shift the focus to genetic factors important in the development of atopic disease, using a novel mouse strain deficient in eotaxin-3. Together, the results presented here represent a step toward better understanding immune development.
